Mike Ashley is a billionaire businessman and owner of Newcastle United Football Club. News outlets reported in 2008 that he visited a private Mayfair casino and threw down a complex roulette bet worth nearly half a million pounds.

In one spin, he wagered a total of £480,000 on every possible inside wager that includes the number 17. This includes a straight-up bet on 17, all the splits that include the number 17, all the streets, corners and six-line bets. This is called a full complete.

The same news reports also state that he placed wagers on black, odd, the first half, middle column and middle dozen. In the end, he took home roughly £1,300,000 for a net profit of £820,000. It’s not a whole lot of money for a billionaire, but it is impressive that the ball landed exactly where it needed to land to make the bet a perfect winner.

Joseph Jagger worked in the cotton mechanics industry and took his experience with machines to the Beaux-Arts casino in Monte Carlo. There, he paid six clerks to secretly note the results of many spins of the roulette wheels there. He found that one of the wheels showed a clear bias which resulted in the numbers 7, 8, 9, 17, 18, 19, 22, 28 and 29 hitting more often than they should have in a truly random wheel.

Joseph Jagger then visited the casino himself, placed his bets accordingly and won the equivalent of $375,000 over the next few days. The casino eventually wised up to his strategy and got rid of the biased wheel. Jagger kept his winnings, retired from his job, invested the money in his own property and never again returned to the casino.

To put that in perspective, $375,000 back then would have a purchasing power of more than $7,000,000 today. The exact numbers are actually different everywhere you look, but $375,000 seems to be the most frequently cited number. Whatever it was, it was a lot of money.

But No-Limit isn't the only way to make bets in poker. In fact for decades the most commonly played forms of poker were slow, steady 'Limit' betting rounds that kept variance and wild bankroll swings to a minimum. Pot-Limit formats (more on this below) are also quite common (eg Pot-Limit Omaha).
In this beginners guide to poker betting we'll take a look at the most common betting rules in Texas Hold'em and beyond. We'll start with the most popular one, of course - No Limit. It's easier to explain, even though it's not at all easy to master.
Beginners Guide to Poker Betting
No-Limit Poker
In No-Limit Poker, as soon as it's your turn to bet you're allowed to bet all the chips that you have in front of you into the pot. You don't even have to have the most chips at the the table -- you can go 'all in' with whatever you have in your stack.
As we mentioned it makes for great drama at the table and tense, cards exposed Texas Hold'em showdowns where one player is playing for their cash game or tournament life on the turn of a single card.
Don't get confused by the exaggerated scenarios of film or TV though - you still can't throw your car keys or your bearer bonds into the pot as they do it in the movies. You can't even dig into your wallet for more cash in the middle of a hand.

Today's No-Limit poker games always use a rule called 'table stakes.' It means that you can never bet anything above and beyond the money you had on the table when the hand started.
As the sharp observer will have noticed this means that there's a 'limit' to the betting after all. So 'no-limit' poker isn't actually without limits. But for the sake of simplicity, No Limit is the term used to describe it.
Don't make the mistake of thinking that no-limit poker is more dangerous for your bankroll than fixed-limit poker. It all depends on what stakes you play at. A game of Limit Texas Hold'em with blinds of $100/$200 certainly isn't cheaper than a No-Limit Texas Hold'em game with blinds of $1/$2.
Fixed-Limit Poker
In fixed-limit poker, the size of each bet is fixed in advance. In Hold'em and Omaha, the first two betting rounds use bets and raises the size of the big blind (called the small bet). In the following two betting rounds, bets and raises are twice the big blind (called the big bet).
When you specify the size of a fixed-limit game, the convention is to give the size of the small bet and the big bet. If the blinds are $1/$2, you'd say that the game is $2/$4. For the internet generation this may seem a bit odd, and it's different from no-limit and pot-limit poker. Still, it's common use.
Often, the number of raises in each betting round is limited to three or four, after which the betting is 'capped.' This means that you won't be able to put in more than $6 or $8 during the first round of betting in a Texas Hold'em game with blinds at $1/$2.
This rule is often put out of play when only two players remain in the hand, in which case they can continue raising until all their money is in the pot. If they want to, that is.
Don't make the mistake of thinking that fixed-limit poker is easier than no-limit poker. Sure, you don't stand to lose your entire stack after a single mistake, but on the other hand you won't double your stack in one single move either. Fixed-limit is another game altogether and you have to play it differently.
Pot-Limit Poker
In Pot-Limit poker the amount you can bet when it's your turn is limited by the size of the pot. The pot-limit rule goes like this:
You can raise up to the amount that is in the pot after you have called the previous bet.

Don't make the mistake of thinking that pot-limit poker is safer for your bankroll than no-limit poker. Even if they are limited to the size of the pot, bets in pot-limit poker are generally not smaller than in no-limit.
Most bets in no-limit poker are actually the size of the pot or smaller.
How Betting Rounds Work in Poker
Each poker hand is made up of a number of betting rounds. The number of betting rounds depends on the poker variation.
In Texas Holdem there are four betting rounds. In Seven Card Stud there are five and in Five Card Draw there are just two betting rounds.
Fold, Call or Raise
In each betting round, the betting moves clockwise around the table. Each player in turn must either match the bet of the previous player (call) or get out of the hand (fold).
Or, instead of just calling, when it's your turn to bet you can also choose to bet more than the previous bet (raise).
When all players have either folded or called the last raise, the betting round is over. All bets that have been made during the betting round are added to the pot.
All players who remain in the hand have now put in the same amount. They have all matched the biggest bet in that betting round. You can think of this as a negotiation - players agreeing on the price to see another card.
When the betting round is over, if all players except one have folded, the remaining player wins the pot. If everybody else but you folds, you don't even have to show your cards to win. That's what makes bluffing possible in poker.
The Check
Before a bet has been made in the current betting round, the player whose turn it is can choose not to bet (check). Checking simply means passing on the turn to the next player without making a bet.
If it helps, you can think of checking as calling a zero bet. It it doesn't help you, please just forget about it.
The Check-Raise
Let's say that a player checks and another player puts in a bet. When the betting comes around to the player who checked may either fold, call the additional but - or raise!
If he raises here his move is called a 'check-raise.' This is not really a rule per se but it's still good to know what check-raising means.
Texas Hold'em Betting Order & The Blinds
At the start of each poker hand some players have to make a bet even before the cards are dealt.

you should get crafting slots as you level up in crafting. you get one after you finish the first quest in wizard city you get one in kt first quest and you get third between mb and ds. hope this helps

To be honest at the max crafting rank we should have 10 crafting slots its not right that we still only have 3 were high rank crafters and when i make something in real life i dont have to wait for invisible crafting timers to cool down to make 10 of something but usually around then i run out of materials.

In real life you cannot possibly make a Hat, Robe and Boots in a matter of seconds like you can in the game. The cool down times and slots are reasonable. Nothing needs more than a day, and be realistic, how many things in real life can you possibly craft in a day. Go make a basic plain silver ring in a day in real life and then come back and tell us you can do 10 things instantly at once without a cool down time.

Crafting slots open up as you complete the quests in each world. The cool down also becomes less of a problem. Hang in there it's gets better.
By the time I finished the Celestia Seal of Seven Seas I was able to craft the Wintertusk gear for all my other wizards at a pretty reasonable rate.

Methods[edit]
A general dot blot protocol involves spotting 1-2 microliters of a samples onto a nitrocellulose or PVDF membrane and letting it air dry. Samples can be in the form of tissue culture supernatants, blood serum, cell extracts, or other preparations.^[3]
The membrane is incubated in blocking buffer to prevent non-specific binding. It is then incubated with a primary antibody followed by a detection antibody or a primary antibody conjugated to a detection molecule (commonly HRP or alkaline phosphatase). After antibody binding, the membrane is incubated with a chemiluminescent substrate and imaged.
Apparatus[edit]
Dot blot is conventionally performed on a piece of nitrocellulose membrane or PVDF membrane. After the protein samples are spotted onto the membrane, the membrane is placed in a plastic container and sequentially incubated in blocking buffer, antibody solutions, or rinsing buffer on shaker. Finally, for chemiluminescence imaging, the piece of membrane need to be wrapped in a transparent plastic film filled with enzyme substrate.
Vacuum-assisted dot blot apparatus has been used to facilitate the rinsing and incubating process by using vacuum to extract the solution from underneath the membrane, which is assembled in between several layers of plates to ensure good seal between sample wells, hold waste solution, and deliver suction force. For chemiluminescence signal detection, apparatus need to be dissembled and the membrane need to be taken out and wrapped in a transparent plastic film.

ABSTRACT
Lacking access to an affordable method of high throughput immunoblot analysis for daily use remains a big challenge for scientists worldwide. We proposed here Quantitative Dot Blot analysis (QDB) to meet this demand. With the defined linear range, QDB analysis fundamentally transforms traditional immunoblot method into a true quantitative assay. Its convenience in analyzing large number of samples also enables bench scientists to examine protein expression levels from multiple parameters. In addition, the small amount of sample lysates needed for analysis means significant saving in research sources and efforts. This method was evaluated at both cellular and tissue levels with unexpected observations otherwise would be hard to achieve using conventional immunoblot methods like Western blot analysis. Using QDB technique, we were able to observed an age-dependent significant alteration of CAPG protein expression level in TRAMP mice. We believe that the adoption of QDB analysis would have immediate impact on biological and biomedical research to provide much needed high-throughput information at protein level in this “Big Data” era.

INTRODUCTION
Lacking of an accessible high throughput immun-oblot method significantly hinders any attempts to investigate the molecular basis of biological and pathological processes systematically in an average research lab. Until now, Western blot analysis remains the most commonly used immunoblot tool in the basic research lab almost 40 years since its invention [1-4]. However, its limitations, including complicated processing steps, limited ability to process many samples, ambiguity in result analysis and requirement of large amount of total protein lysate for analysis, determines this technique an unlike choice for high throughput analysis. As an alternative, Dot blot analysis was developed to simplify the process of Western blot analysis [5]. In fact, both enzyme-linked immunosorbent assay (ELISA) [6-8] and reverse phase protein microarray (RPPM) [9, 10] can be considered as Dot Blot analysis in a high throughput format. Nonetheless, the applications of these techniques are still limited in basic research lab for various reasons, including limited availability for prefabricated ELISA kits and lack of easy access to RPPMs.
Meanwhile, with the rapid development of effective high-throughput tools in genetic research, there are strong demands for complementary high throughput immunoblot methods on a daily basis for biomarker identification and other association studies at protein level [11, 12]. Ideally, the basic strategy shows the play which, on average, will result in the greatest win or the least loss per initial hand played. The Best Starting Hands in Hold’em
Let’s start by talking about the best starting hands, which are often referred to as ‘premium hands’. There is some disagreement amongst poker players as to which starting hands are the best, but few would dispute the value of the first of our three main groups, Aces and Kings.
Group 1: AA, KK
These two starting hands are the major players in hold’em. It’s not often you’ll get dealt Aces or Kings. In fact you get either Aces or Kings once in every 110 hands, so it’s not nearly as often as we’d like. Aces are by far the best possible starting hand in hold’em, closely followed by Kings. However, you should be aware that even Aces or Kings can get cracked, and they don’t play too well against multiple opponents. This means you should definitely be raising pre-flop to narrow the field. Extra caution is necessary when playing Kings, because if an Ace falls on the flop then you’re losing to anyone who has a single Ace in their starting hand. While they are very strong hands which most players love to get, they are certainly not unbeatable.
Group 2: QQ, JJ, AKs
Queens and Jacks are great starting hands, and with either of these, you can usually be confident you have the best starting hand. Of course they are dominated by Aces and Kings, but they’re a favourite against all other starting hands. While Queens and Jacks will occasionally run into a player holding either Aces or Kings, it doesn’t happen too often. Play these cards strongly, and always look to raise with them.
Ace-King is known throughout the poker world as Big Slick, and when suited it’s often called Super Slick. While it isn’t a ‘made hand’, unlike a pair, it offers great potential. It’s only a big underdog to Aces and Kings, and even pairs like Queens and Jacks are only slight favourites. The beauty of AK (suited or unsuited), is that it dominates so many other hands like AQ, AJ, AT, and so on. These types of hands are the ones that players usually end up pushing all-in with late in a tournament.
Group 3: TT, AK, AQs, AJs, KQs
This next group of starting hands is also a strong bunch. You should definitely be looking to raise pre-flop with any of these hands too. We’ve already talked about the power of AK, but starting hands like AQs, and AJs, are also very strong and often run into weaker Ace-X combinations. Even though these are all strong starting hands, and most of the time you’ll be winning pre-flop, you have to be careful – particularly a hand like KQs, which you can easily fold to a re-raise.
Suited Cards
You’ll often hear novice players responding to questioning of why they played a particular starting hand with the line “well, because they were suited”. Some suited cards are worth playing and it’s certainly better to start with suited cards than unsuited cards. However, the odds of flopping a flush is 1 out of 118 hands (0.8%) with two suited cards, and you’ll only make a flush after the river around 6.5% of the time. Don’t fall into the trap of playing any two cards just because they happen to be suited – it doesn’t make a big enough difference to make junk hands valuable.
Kicker Issues
The word ‘kicker’ means the smaller of your two cards. Some players play a hand if it contains an Ace with any other card (such as an Ace with a 3 kicker), and this type of play ultimately cost players money and tournaments. For example, let’s suppose a player calls with A6 and the flop comes A83. What does the player do? bet? call? raise? call a big raise? go all-in? What if the flop comes Q63? The player has middle pair – which is very hard to play. Hey, the flop could come A6X – the player has two pair, Aces and sixes but this happens only 1 out of 49 hands (2%). Until you learn when and how to play Ace junk (AX) go slow with it. One good thing about A junk and K junk, is that you do not need to play these hands to learn when they may be profitable. Hold’em starting hands can be a complex subject because every situation is different. If you were to ask a professional poker player, “should I call, raise, or fold this hand pre-flop?” his response would almost certainly be “it depends!” Here are some of the main reasons why it depends:
The Number of Players
The value of certain starting hands is very dependent upon the number of players at the table. Certain starting hands are always going to be under threat against a table of nine or ten players, but the value of these same hands increases when there are fewer players. A starting hand like KJ might be vulnerable against a full table of players, but is considered a strong hand if there are just a few other players.
Position
Your position on the poker table will be a major factor in deciding which starting hands you should play. The later your position in the betting order, the better – because you get to decide what to do after most of your opponents have acted. We’ll talk much more about the importance of position throughout our lessons on Pokerology, but as a first step please see our lesson on the value of position. Playing position can elude us at first because it is a part of poker that lends itself to be exploited through experience. However, you must quickly realize that your position at the table should heavily influence the choice of starting hands that you play. Until a player has a feel or grasp for positional play, just believe and follow some of the suggestions on the subject.
A Raised Pot
Whether or not a pot has been raised should be a very important factor in your decision to play a particular starting hand. Your selection of starting hands should change when the pot has been raised by a reasonable player. If there has been a raise and a re-raise before you’re due to act, then you should only consider playing with a very strong hand. Of course this will also depend on the personality types of the other players and whether the game is very loose or passive.

Interac e-Transfer (formerly Interac Email Money Transfer) is a funds transfer service between personal and business accounts at participating Canadian banks and other financial institutions, offered through Interac Corporation.
From inception until early 2018, the service was provided by Acxsys, a for-profit consortium backed by most of the major partners of the nonprofit Interac Association, and using the Interac brand under licence. In February 2018, the activities of both organizations were combined into a single for-profit organization under the Interac name.^[1]
Participating institutions[edit]
Most Canadians who use online banking can send funds. These include personal deposit account holders with the big five banks, Desjardins, Tangerine, National Bank, President's Choice Financial, and many credit unions and other institutions,^[2] as well as some small-business account holders.^[3] In 2015, 105 million money transfers were sent using the platform totaling over CA$44 billion in value.^[4]
Any personal account holder in Canada can receive funds.
How it works[edit]
An e-Transfer resembles an e-check in many respects. The money is not actually transferred by e-mail. Only the instructions to retrieve the funds are.
The sender opens an online banking session and chooses the recipient, the amount to send, as well as a security question and answer. The funds are debited instantly, usually for a surcharge. The sender sends the security answer separately to the recipient, usually through another medium outside of the e-mail as a secondary security measure.
An e-mail or text message is then sent to the recipient, with instructions on how to retrieve the funds and answer the question, via a secure website.
The recipient must answer the security question correctly. (If the recipient fails to answer the question correctly after three tries, then the funds will automatically be returned to sender.^[5])
If the recipient is subscribed to online banking at one of the participating institutions, the funds are deposited instantly at no extra charge.
If the recipient's deposit account is not at one of the participating institutions or not subscribed to online banking at all, the funds are deposited within three to five business days, and a surcharge (currently $4.00) is deducted from the amount received.
The Autodeposit feature allows senders to send money and be received by the recipient without the recipient having to answer a security question. The recipient must enable this feature in their account settings. Not all banks offer this feature.
When an eTransfer has not been accepted after a certain period of time the transfer will not go through. The transfer duration depends on bank and/or on the persons settings. Some eTransfers can be automatically canceled after 24 hours or for a period of up to 30 days depending on bank/user. Banks like TD, CIBC and RBC have a set 30 day limit until an eTransfer is cancelled, while other banks have shorter durations and set limits.
Benefits and disadvantages[edit]
Unlike a cheque, the funds from an e-Transfer are not frozen in the recipient's account. An e-Transfer cannot bounce, as the funds are guaranteed, having been debited from the sender's account immediately upon initiating the transfer. As long as both sender and recipient bank are participating institutions, the funds are sent and received instantly. However in some cases, for example two people with different banking institutions, transfers may take anywhere between near instant, or up to a few hours for the receiving party to get their emailed notice.
However, like any online banking mode of payment, e-Transfers are vulnerable to phishing. Many Canadians in areas where the Big Five banks have little presence or who do not bank online are penalized by a surcharge when receiving e-Transfers. Unlike a real giro, an e-Transfer requires intervention from the recipient for every single transaction unless the recipient has signed up for Direct Deposit. An e-Transfer goes stale much faster than a cheque (after 30 days, the e-Transfer is automatically cancelled and the sender is notified by e-mail to retrieve the funds).^[6]
